Greifswald - Malmö          2 Autostunden + 3,5 Stunden Fähre ab Mukran
Greifswald - Stettin          120 km
Greifswald - Insel Rügen    30 km
Greifswald - Insel Usedom 30 km

Straßenanbindung
Über die Ostseeautobahn A 20 Lübeck-Stettin ist die Universitäts- und Hansestadt Greifswald mit den benachbarten  Metropolregionen Berlin, Hamburg und Stettin verbunden.
Die AS (Anschlussstelle) Greifswald befindet sich 12 km westlich und die AS Gützkow/Greifswald 18 km südlich vom Stadtzentrum.
Hauptverkehrsachse nach Norden zur Insel Rügen und den skandinavischen sowie baltischen Staaten ist die B105 auf den Rügenzubringer B 96 (über die A 20). Zur Insel Usedom und in den Raum Anklam/Pasewalk übernimmt die B 109 diese Funktion. Die beiden  Bundesstraßen 105 und 109 bilden zudem südlich und westlich der Stadt eine leistungsfähige Ortsumgehung.
Der Ortsteil Greifswald-Insel Riems  befindet sich zirka 10 km nördlich des Stadtzentrums. Die Abfahrt von der B 105 zur Insel Riems ist ausgeschildert.

Bahnanbindung
Die Universitäts- und Hansestadt Greifswald liegt an der Bahnstrecke Stralsund/Sassnitz-Pasewalk-Berlin. Die Strecke wird gegenwärtig für Reisegeschwindigkeiten bis 160 km/h ausgebaut.
Fernverkehrsverbindungen mit dem InterCity bestehen

in Richtung Berlin Hbf (und weiter bis Halle/Erfurt sowie einmal täglich bis Prag)
in Richtung Hamburg Hbf (mit Umstieg in Stralsund)

einzelne Direktverbindungen bestehen weiterhin

von Greifswald über Hamburg, Köln, Stuttgart (Montag-Freitag)
nach Hamburg bzw. Köln (an den Wochenenden)

Zusätzlich stehen Direktverbindungen mit der Usedomer Bäderbahn (UBB) von der Universitäts- und Hansestadt Greifswald nach Osten über die Insel Usedom bis  Polen (Swinemünde) sowie Verbindungen nach Norden zur Halbinsel Fischland/Darß bis Barth (mit Umstieg in Stralsund) zur Verfügung.
Über einen Gleisanschluss ist der Stadthafen Greifswald-Ladebow mit dem Schienennetz der Bahn verbunden.

Die Universitäts- und Hansestadt Greifswald  liegt direkt am Greifswalder Bodden/Ostsee. Sie bietet damit für den Schiffsverkehr und den Wassersport günstige natürliche Bedingungen.
Über eine Direktansteuerung von See aus ist der Stadthafen Greifswald-Ladebow zu erreichen. Die Fahrrinne ist 6,9 m tief. Die straßenseitigen Anbindungen sind bis zur Ostseeautobahn A 20 für Schwerlasttransporte ausgebaut. Der Stadthafen hat außerdem einen Gleisanschluss.
Mit dem deutschen und polnischen Binnenwasserstraßennetz ist die Universitäts- und Hansestadt Greifswald direkt über die Oder verbunden.
Das Stadtzentrum mit seinem einzigartigen Museumshafen liegt 4 km landeinwärts. Es ist über den schiffbaren Fluß Ryck mit seiner 4 m tiefen Fahrrinne gut erreichbar. Entlang des Flusses befinden sich die Sportbootliegeplätze und das maritime Gewerbe, unter anderem auch die Produktionsstätte der HanseYachts AG.
Sporthäfen befinden sich in Greifswald-Wieck und in der Salinenstraße in Greifswald. Der Museumshafen befindet sich in der Altstadt.
